EGF Family Receptors

  • EGF family of receptors are transmembrane RTKs that are frequently overexpressed or mutated in a wide variety of epithelial tumors.
  • The EGF or ERBB family consists of EGFR (ERBB1), HER2 (ERBB2), HER3 (ERBB3), and HER4 (ERBB4).
  • They are characterized by an extracellular ligand-binding domain and an intracellular tyrosine kinase domain except ERBB3, which has a kinase-deficient intracellular domain.

Activation of the Receptors

  • The EGF family of receptors is activated by binding to a ligand resulting in homo- or hetero-dimerization of the receptors and phosphorylation of the receptors at the recruitment of adapter molecules, leading to activation of signaling pathway cascades downstream.
